摘 要:目的建立改良的Caco-2细胞模型研究丹参中的可吸收成分,并用在体动物模型验证。方法HPLC-MS方法测定加入丹参水提取物的Caco-2细胞破碎液中的丹参成分,并与大鼠在体灌流给药后血浆中的成分相比较。结果加药Caco-2细胞破碎液中检测到丹参素、原儿茶醛、咖啡酸、丹酚酸D、紫草酸、迷迭香酸、丹酚酸B7种丹参的成分,给药后大鼠血浆中检测到丹参素、原儿茶醛、咖啡酸、丹酚酸D、紫草酸、迷迭香酸、丹酚酸B、丹酚酸A共8种丹参的成分。结论细胞破碎液中检测的成分为丹参在Caco-2细胞中有吸收的成分,本方法可作为研究中药吸收的新方法。AIM: To study the constituents of Salviae miltiorrhizae absorbed by improved Caco-2 cell model. METHODS: Using HPLC/MS analysis, the water-soluble constituents in Radix Salviae miltiorrhizae absorbed by Caco-2 cell were analyzed and identified, in comparison with the constituents in plasma of rats following administration. RESULTS: Seven constituents absorbed in Caco-2 cell were identified as Danshensu, protocatechuic aldehyde, caffeic acid, salvianolic acid D, lithospermic acid, rosmarinic acid and salvianolic acid B by LC-EST/TOF MS. Eight constituents in plasma of rats following administration danshensu, protocatechuic aldehyde, caffeic acid, salvianolic acid D, lithospermic acid, rosmarinic acid, salvianolic acid B and salvianolic acid A were detected. CONCLUSION: The developed method could simply and effectively estimate the absorption of traditional Chinese medicine in vivo.
