摘 要:观察了米力农(Mil),lemakalim(Lem)和维拉帕米(Ver)三种血管舒张剂对前列腺素F2α(PGF2α)所致缺氧2和4wk大鼠肺动脉和主动脉环收缩反应的影响.Lem(0.01~1.0μmol·L-1),Mil和Ver(均为0.1~10.0μmol·L-1)对主动脉和肺动脉都有明显的抑制作用,使PGF2α引起的血管收缩反应曲线压低,EC50显著增高(P<0.001),呈剂量相关关系.其抑制强度为Lem>Mil>Ver,缺氧前后顺序不变.无论缺氧前后,Lem和Ver对主动脉和肺动脉的抑制强度都较接近,Mil则在缺氧条件下。The effects of milrinone (Mil), lemakalim [Lem, ( ) 6 cyano 3,4 dihydro 2,2 dimethyl trans 4 (2 oxo 1 pyrrolidinyl) 2H benzo pyran 3 ol] and verapamil (Ver) against contraction of pulmonary artery ring (PAR) and aorta ring (AR) from chronically hypoxic rats (hypoxia 2 and 4 weeks) induced by prostaglandin F 2α (PGF 2α ) were studied in vitro . Lem (0.01 1.0 μmol·L 1 ), Mil (0.1 10.0 μmol·L -1 ) and Ver (0.1-10.0 μmol·L -1 ) could remarkably inhibit vasocontraction in both of AR and PAR, with the depression of contractile responses elicited by PGF 2α and obvious increase in EC 50 , in a dose dependent manner. Among three vasodilators the order of inhibitory strength was Lem>Mil>Ver and this order never changed whether in normoxic rats or in hypoxic rats. Furthermore, the inhibition strengths of Lem and Ver against contraction of PAR and AR induced by PGF 2α were similar in values, either before or after hypoxia. However, the inhibitory strength of Mil was selectively increased in chronically hypoxic PAR.
