摘 要:用20只SD大鼠的坐骨神经片段培养在涂有层粘蛋白的聚酯纤维上,对照组培养在无层粘蛋白的聚酯纤维上.培液由20%小牛血清和80%DMEM组成,37℃,5%二氧化碳培养箱培养2周.在培养24,48,72小时,观察雪旺细胞迁移情况,测量迁移最远的雪旺细胞离组织块边缘的距离.结果显示:实验组和对照组相比较,雪旺细胞从组织中迁出的时间早.速度快,数量多.在体外培养条件下.显示了层粘蛋白对雪旺细胞在三维网架上迁移的促进作用.对层粘蛋白促进雪旺细胞迁移的影响和作用机制作了讨论,并探讨其在神经移植中的应用前景.The sciatic nerve segments , obtained from newborn Sprague-Dawley rats and dissected under sterilized condition were cultured on the polyester fibers coated with laminin as the experimental group and on the polyester fibers without laminin as the control group. All the segments were incubated in DMEM containing 20% calf serum and maintained in 5% CO2 at 37 ℃ for two weeks. The migration of Schwann cells from the segments was observed and the farthest distance of the migration from the segments was measured at 24. 48 and 72 hours after incubation. The results were as follows:The migration time of Schwann cells occured earlier in the experimental group, the number of Schwann cells greatly increased and the velocity of Schwann cell's migration was greater. The effects of laminin upon the migration of Schwann cells on the three-dimentional f'ramwork in vitro, the mechanism of laminin, and the prospect of the application of laminin in nerve transplantation were discussed.
