添加营养物质实现1-苯基-1,2-乙二醇的高效转化

摘  要:为提高近平滑假丝酵母多批次转化(S)-1-苯基-1,2-乙二醇(PED)的对映体过量值(ee值)、转化率和多批次转化的稳定性,研究不同营养物质对去消旋反应转化(S)-PED的影响。结果表明:在添加的营养物质中,牛肉膏与氯化铵同时添加效果最好,其ee值和转化率达到95.87%和96.17%,比不添加的分别提高18.45%和24%;当添加5 g·L^(-1)乙醇或乙酸钠时,在不降低转化效果的同时反应批次由原来的2次提高到8次;通过胞内酶活性分析发现:添加乙醇能够激活乙醛脱氢酶的同时,使乙醇脱氢酶、6-磷酸葡萄糖脱氢酶、羰基还原酶的活性分别提高54.54%、42.01%、173.57%。Several nutrition were investigated for improving the ability to convert (S)-1-pheyl-1,2-ethanediol (PED) by Candida parapsilosis through deracemization. The results showed that complex nutrition were better than carbon source and nitrogen source. And nitrogen source can increase the value of yield of product. The value of enatiomeric excess (ee) and yield increased to 95.87% and 96. 17% after beef extract and ammonium chloride were added at the same time into reaction solution. The batch cycles increased from 2 to 8 when alcohol or acetate sodium was added by acitvation of the enzymes involved in the deracemization reaction and coenzyme-regeneration reaction such as aldehyde dehydrogenase,alcohol dehydrogenase,glucose-6-phosphate dehydrogenase and carbonyl reductase activity.
