摘  要:目的:改良99Tcm-WBC制备条件并评价其特性.方法:取10ml家兔及人全血分离WBC,用99Tcm-HMPAO介导标记,评价WBC分离率、99Tcm-WBC的标记率、稳定性、活性及其在家兔脓肿模型和炎性肠病患者体内的炎症定位特性.结果:WBC分离率达70%,99Tcm-WBC标记率和活性率分别高达60%和90%.放射性不断自WBC中逸脱.注射99Tcm-WBC后10min家兔大腿脓肿即显影,4h时最清晰.99Tcm-WBC可清晰地显示活动性炎性肠病受累肠段部位和范围.结论:10ml血可分离和制备出满足临床显像要求的99Tcm-WBC.Objective: To improve 99 Tc m WBC preparation conditions and assess its properties for focal inflammation detection. Methods: Rabbit and human WBC were separated from 10 ml whole blood and labeled with 99 Tc musing 99 Tc m HMPAO. 99 Tc m WBC was evaluated in labeling efficiency, stability and viability. The property of 99 Tc m WBC in focal inflammation localization was evaluated in a rabbit abscess model and three patients with active inflammatory bowel disease (IBD). Results: The separation yield reach up to 70%, and 99 Tc m WBC labeling efficiency and viability were 60% and 90% respectively. Radioactivity eluted continuously from 99 Tc m WBC when in vitro incubated. 99 Tc m WBC accumulated in the rabbit abscess as early as 10 min post injection (p.i.) and culminated at 4 h p.i.. 99 Tc m WBC could definitely elucidated the inflammatory foci in patients with active IBD. No sideeffects were observed p.i. of the radiopharmaceutical. Conclusion: 10 ml whole blood could prepared desirable 99 Tc m WBC for inflammation detection.
