摘  要:目的:检测蛇毒多肽(卡迪,KD)和蝎毒多肽(polypeptide from Buthus martensii venom,PBMV)对肝癌移植瘤(H22)带瘤小鼠瘤重的抑制作用,并观察肿瘤细胞Caspase-3表达,初步探讨其作用机理。方法:昆明种系近交小白鼠,60只,复制肝癌移植瘤模型,随机分为6组:阴性对照组(生理盐水),阳性对照组(环磷酰胺0.010mg/mL),KD0.016、0.010mg/mL组和PBMV0.016、0.010mg/mL组,分别注射KD、PBMV和阳性药物10d后,分析统计带瘤小鼠的体重变化、脾指数和肿瘤生长抑制率;并采用免疫组织化学方法,观察治疗后Caspase-3表达的改变。结果:KD0.016、0.010mg/mL组和PBMV0.016、0.010mg/mL组的平均瘤重显著地减小,均具有明显的肿瘤生长抑制率,与阴性对照组相比有显著差异(P<0.05),而与阳性对照组相比没有显著差别(P>0.05)。免疫组化显示:阴性对照组胞浆无着色或着色很浅;阳性对照组胞浆浅红棕色,着色胞浆分布较集中;两组待测组胞浆均显红棕色,分布面积广,而且随着用药浓度的增加而增多。结论:KD和PBMV对H22生长和增殖具有抑制作用,其抑制作用的机制可能是通过增强Caspase-3蛋白的表达而促使肿瘤细胞的凋亡。Objective:To explore the effects of snake venom polypeptide (KD) and polypeptide from Buthus martensii venom (PBMV) on tumor weight and the expression of Caspase-3 in Hepatoma 22-bearing mice. Methods: Sixty H22-bearing mice were divided into 6 groups in random: negative groups (N. S), positive groups (CTX), KD 0. 016,0.010 mg/mL groaps and PBMV 0. 016,0. 010 mg/mL group. After dosage of KD, PBMV and positive control for 10 days,the weight of H22-bearing mice,spleen weight index and tumor growth inhibition rate (IR%) were observed. By using immunohis-tochemistry method, expression of Caspase-3 were studied. Results: The mean tumor weight of KD groups and PBMV groups decreased significantly and tumor growth inhibiting rate was significantly high when compared with negative control(P 〈 0. 05 ), but there were not significantly different from those in positive control ( P 〉 0. 05 ). Immunohistochemistry showed:Caspase-3 protein was achromic or hypochromic in the cytoplasm in negative groups, while positive groups appeared red-brownish with concentrated stained cytoplasma. The treatment groups showed strong red-brownish cytoplasm in a wide range of distribution,which increased with ascending doses. Conclusion: The growth of H22 can be inhibited by KD and PBMV. By inducing the express of Caspase-3 protein and promote the tumor cell apoptosis may be the mechanism of the inhibitory effect of KD and PBMV.
