摘  要:探讨新型药物FK520的免疫抑制功能。以ConA非特异性多克隆活化淋巴细胞,研究FK520对该增殖的免疫抑制效应;用尼龙毛柱分离OVA(卵清蛋白)TCR转基因小鼠T淋巴细胞后,以OVA323-339肽致敏的抗原提呈细胞刺激该T淋巴细胞增殖,研究FK520对抗原特异性T淋巴细胞增殖的免疫抑制效应;将20只5周龄NOD小鼠随机分为两组,每周分别给NOD小鼠肌肉注射FK520或溶剂2次,每次给予FK520药物10 mg/kg或相同体积溶剂,观察FK520对NOD小鼠体重、血糖及胰岛炎的影响。结果显示,FK520体外不仅能显著抑制淋巴细胞非特异性多克隆活化增殖,而且能抑制抗原特异性T淋巴细胞增殖,该免疫抑制效应具有量效依赖关系。体内注射FK520能显著降低NOD小鼠IDDM发病率(P<0.05),推迟小鼠IDDM的发生。上述结果表明,FK520具有显著的免疫抑制效应和潜在对自身免疫性疾病的治疗作用。To investigate the immunosuppressive effects of FK520, splenocytes from BALB/c mice were stimulated with ConA, and T lymphocytes derived from DO 11.10 transgenic mice were enriched by nylon wool and then incubated with OVA323-339 peptide plus APC. Separately, 5 weeks old nonobese diabetic (NOD) mice (n=20) were divided into two groups randomly and immunized by intramuscular injection with FK520 (10 mg/kg) or with an equal volume of the dissolvent twice a week. The effects of FK520 on the body weight, blood sugar and insulitis of NOD mice were observed. The proliferations induced either by ConA or by specific antigen peptides could be inhibited by FK520 in a dose dependent manner. In addition, the IDDM incidence of NOD mice was reduced significantly by FK520 (P〈0.05), which delayed the onset of the disease. From the observations in the present study, it is evident that FK520 shows a potent immunosuppressive effect and can be used as a potential therapeutic agent for autoimmune diseases.
