摘 要:目的研究心梗后心力衰竭(心衰)大鼠的海马区基因表达变化特征。方法采用结扎左冠状动脉前降支的心梗模型,利用基因表达谱芯片技术,研究大鼠心衰10 d和心衰60 d,海马区和皮质区的基因表达变化。结果心衰10 d大脑的基因变化明显比心衰60 d时活跃,心衰10 d时海马区的阳性差异基因为24条,而心衰60 d时为14条,主要涉及到的有能量代谢、神经递质代谢、神经鞘脂、信号转导、应激和免疫反应等方面作用的基因。其中,能量代谢类基因丙酮酸激酶3(NM_053297)和3-羟3-甲基辅酶A裂解酶(NM_024386)基因在心衰10 d时下调;神经递质代谢类基因烟碱型ε乙酰胆碱能受体(NM_017194)、alpha Endosulfine(NM_021842)和谷氨酸草酰乙酸转氨酶2(NM_013177)基因在心衰10 d时下调。结论心衰初期大脑海马区的能量代谢类基因和神经递质代谢类基因发生了变化。Objective To study the change characteristics of gene expression of hippocampus in rats with congestive heart failure after heart infarction. Method The model of heart infarction was established by ligation of left front descending coronary artery and computer chip technique of gene spectrum was used to study the changes of gene expressions in the hippocampus and cortex in rats with congestive heart failure for 10 days and for 60 days respectively. Result The changes of gene expression in 10-day group was more active than those in 60-day group. There were 24 genes in hippocampus in 10-day group and 14 genes in 60-day group. These genes were mainly related to genes of energy metabolism, neurotransmitters metabolism, sphingoin, signal transduction, stress and immune reaction. Among these genes the expressions of energy metabolism genes, such as pyruvate kinase 3 (NM_053297) and HMG-CoA lyase (NM_024386) were decreased from 10th day of congestive heart failure, and the expressions of neurotransmitters metabolism genes, such as cholinergic receptor (nicotinic, epsilon polypeptide, NM_017194), endosulfine alpha (NM_ 021842 ) and glutamate oxaloacetate transaminase 2 (NM_013177) were decreased from 10th day of congestive heart failure. Conclusion The genes of energy metabolism and neurotransmitters metabolism were changed in brain hippocampus in primary congestive heart failure.
