摘 要:目的观察复方瑞康欣对吗啡依赖人神经母细胞瘤SK-N-SH细胞[Ca2+]i的影响并探讨其机制。方法SK-N-SH细胞周期同步化后,用10μmol/L的吗啡作用48 h,建立吗啡依赖细胞模型;再用复方瑞康欣含药血清干预12 h,纳洛酮戒断,Fluo 3-AM标记后,流式细胞仪检测SK-N-SH细胞内游离Ca2+荧光强度。结果复方瑞康欣含药血清中、高剂量组细胞内游离Ca2+荧光强度均降低,与空白血清戒断组比较,有显著性差异(P<0.05)。结论复方瑞康欣的戒毒作用可能与其下调细胞Ca2+水平有关。Objective To observe the influence of Fufang Ruikangxin Granules on [ Ca^2+ ] i of morphine dependent SK-N-SH cell of human neuroblasoma and the mechanism. Method After the cell cycle synchronized SK-N-SH cells were treated with morphine of 10 μmol/L for 48 hours to establish the morphine dependent model. The model was intervened with Fufang Ruikangxin Granules serum for 12 hours, and then abstained by naloxone. The fluorescent intensity of free Ca^2+ in SK-N-SH cells was detected by flow cytometer after the cell stained with Fluo-3-AM. Result The fluorescent intensity of cell free Ca^2+ was decreased in high dose and medium dose groups of Fufang Ruikangxin Granules. The difference was significant compared with the blank group ( P 〈 0. 05 ). Conclusion The abstinence effect of Fufang Ruikangxin Granules may be related to it can decrease the level of free Ca^2+ in cells.
