摘 要:目的:建立天麻中天麻苷的 HPLC-ELSD 测定方法。方法:采用 Symmetry C_(18)色谱柱(3.9 mm×150 mm,5μm),以乙腈-水(2∶98)为流动相,流速0.8 mL·min^(-1);柱温25℃;蒸发光散射检测器的氮气压力2.76×10~5Pa;漂移管温度90℃;雾化室温度48℃。结果:天麻苷浓度在0.06~0.36 mg·mL^(-1)范围内呈线性关系,平均回收率97.7%,RSD 为2.1%,测得天麻中天麻苷的含量为0.89%,与采用 HPLC-DAD 方法测得的天麻苷含量无显著性差异。结论:HPLC-ELSD 方法测定天麻苷含量准确、可靠,回收率稳定,重复性好,可用于天麻药材及制剂中天麻苷的含量测定。Objective:To establish HPLC - ELSD method for determination of gastrodin in Rhizoma Gastrodiae. Method:The chromatographic method was carried out on Symmetry Ci8 column(3.9 mm× 150 mm,5μm) using acetonitrile - water( 2:98 ) as mobile phase. The flow rate was 0. 8 mL·min^- 1. The column temperature was 25℃. The pressure of N2 in ELSD was 2. 76 × 10^5 Pa. The temperatures of drift tube and atomizer chamber were 90℃ and 48℃ respectively. Results: Gastrodin calibration curves were linear in the range of 0. 06 - 0. 36 mg · mL^-1. The average recovery of gastrodin was 97. 7% with RSD = 2, 1%. The determined content of gastrodin in Rhizoma Gastrodiae was 0. 89%. Conclusion: HPLC - ELSD method for determination of gastrodin is reliable, accurate and have stable recovery and good reproducibility. The method can be used to determine the content of gastrodin in Rhizoma Gastrodiae medicinal material and preparation.
