摘 要:根据已知蚜虫的Buchnera trpB基因序列,设计了1对通用引物,通过PCR扩增法从桃蚜(Myzus persicae(Su lz-er))和豆蚜(Aphis craccivoraKoch)体内扩增出了其内共生菌Buchnera的trpB基因,序列长度约388 bp。通过对桃蚜和豆蚜体内的Buchnera trpB基因序列进行系统发育分析,结果发现桃蚜的Buchnera trpB基因序列与已知的麦双尾蚜(D i-uraphis noxia(Kurd j))的Buchnera trpB基因序列相近,而豆蚜的Buchnera trpB基因序列则与已知的豌豆蚜(Acryrthosi-phon pisum(Harris))的Buchnera trpB基因序列相近。这是桃蚜和豆蚜体内的trpB基因序列的首次报道。Based on the reported trpB gene sequences of Buchnera from other aphid species, a pair of common primers was designed to amplify the trpB gene of Buchnera in Myzus persicae (Sulzer) and Aphis craccivora Koch. The PCR amplification results showed that both of the sequences were about 388 base pairs long. By analyzing the trpB gene sequences of Buchnera in M. persicae and A. craccivora through constituting the phylogenetic tree, it was found that the trpB gene sequence in M. persicae was more closely related to the trpB gene sequence in Diuraphis noxia ( Kurdj), and the trpB gene sequences in A. craecivora and Acryrthosiphon pisum (Harris) had a better similarity relatively. This is the first report on the trpB gene in M. persicae and A. craccivora.
