尿激酶热敏脂质体体内外溶栓效果的研究

摘　　要：目的研究尿激酶热敏脂质体(UKTsL)的体内外溶栓效果。方法在简易体外溶栓装置上研究了UKTsL的体外靶向溶栓效果;在兔颈动脉血栓模型上观察了UKTsL的体内靶向溶栓能力。结果体外溶栓实验显示,UKTsL组的血凝块溶解率达(50.41±4.04)%,和2倍尿激酶(UK)剂量的UK组相近,与磷酸盐缓冲液(PBS)组及尿同UK剂量的尿激酶脂质体(UKL)组相比差异有显著性(P<0.01)。体内实验显示UKTsL组(UK总量75000u/mL)各时段血压变化及血栓形态学改变与UK组(150000u/mL)相近(P>0.05)。结论UKTsL具有较好的热靶向溶栓能力,能减少尿激酶的毒副作用。UKTsL用于溶栓还有待深入研究。[Objective] To investigate the in vivo and in vitro thrombolytic efficacy of urokinase encapsulated thermosensitive liposome UKTsL）. [Method] The simple vitro-thrombolysis-equipment was used to study the targeting thrombolytic efficacy of UKTsL in vitro. Model of carotid thrombosis in rabbit was used to observe the targeting thrombolysis of UKTsL in vivo. [Result] The in vitro thrombolytie experiment demonstrated clot lytic rate of group UKTsL was up to （50.41±4.04）%, which was close to the twice urokinase （UK） dosage of grqup UK. Significant differences （P 〈0.01） were observed between group UKTsL and group phosphate buffer （PBS） and the same UK dosage of group urokinase encapsulated liposome （UKL）. The in-vivo experiment showed that in group UKTsL （total UK 75 000 u/mL） the changes of blood pressure and morphology of thrombus at different time was near to that in group UK （150 000 u/mL） （P 〉0.05）. [Conclusion] UKTsL has the thermal targeting property in thrombolysis, and it can reduce the side effect of urokinase. The thrombolytic efficacy of UKTsL is under deep study.
