摘 要:目的:研究银杏叶片对单侧输尿管梗阻(UUO)大鼠肾脏的保护作用。方法:Wistar大鼠随机分为假手术组、UUO模型组、苯那普利组及银杏叶片组,于给药后2周和4周分别测定各组大鼠肾脏指数、左右肾重比、N-乙酰-β-D-氨基葡萄糖苷酶(NAG)酶活性、尿液尿素氮、尿肌酐、血尿素氮、血肌酐及24 h尿蛋白排泄量等指标。结果:给药后2周,银杏叶组大鼠NAG酶活性[(18.0±5.4)U.L-1]、血尿素氮[(7.3±1.3)mmol.L-1]、血肌酐[(35.2±8.4)μmol.L-1]及24 h尿蛋白排泄量[(9.82±4.42)mg]均明显低于模型组(P<0.05);给药后4周,银杏叶片组大鼠NAG酶活性[(10.3±4.1)U.L-1]、血尿素氮[(8.7±0.9)mol.L-1]、血肌酐[(44.2±6.7)μmol.L-1]及24 h尿蛋白排泄量[(10.17±8.42)mg]均较模型组明显降低(P<0.05);给予银杏叶片能减轻UUO大鼠肾间质纤维化程度。结论:口服银杏叶片对UUO大鼠肾功能和结构具有明显保护作用。Objective To investigate the protective effect of Ginkgo biloba (Gb) tablet on rat kidney following unilateral ureteral obstruction (UUO). Methods Wistar rats were divided into four groups: sham group, UUO model group, Gb tablet treatment group and benazapril treatment group (n= 12). After Gb tablet was given to UUO rats for 2 and 4 weeks, all rats were sacrificed, the index of kidney, ratio of left and right kidney weight, the activity of urinary N acetyl-beta-D-glucosaminidase (NAG), urea nitrogen in urine and serum, creatinine in urine and serum and the 24 h urinary protein excretion were detected, respectively. Results The NAG activity [- (18. 0±5.4) U·L^-1], serum urea nitrogen [ (7.3±1.3) mmol·L^-1, serum creatinine [ (35.2±8. 4) μmol·L^-1 and the 24 h urinary protein excretion [ (9.82±4. 42) mg] in Gb tablet treatment group were decreased significantly compared with UUO model group (P 〈 0.05) 2 weeks after administration. The NAG activity E (10.3±4. 1) U·L^-1, serum urea nitrogen [ (8.7±0.9) mmol ·L^-1], serum creatinine [(44.2±6.7) μmol·L^-1 and the 24 h urinary protein excretion [ (10. 17±8.42) mg] in Gb tablet treatment group were obviously lowered than those in UUO model group after treated with Gb for 4 weeks (P〈0. 05). Gb tablet could lighten the lesion of kidney in UUO rats. Conclusion Gb tablet has obviously protective effect on renal function and structure in UUO rats.
