摘 要:目的:研究新型生物降解可吸收材料聚L-乳酸/聚L-乳酸接枝的羟基磷灰石复合物(PLLA/PLLA-gHAP)的生物相容性,为其应用提供生物学依据。方法:将96孔板上培养的L929细胞分为PLLA/PLLA-gHAP组、PLLA组、阴性对照组(空白培养液)及阳性对照组(含苯酚培养液)4个组,不同组材料浸提液与细胞共培养24、48和72h,以MTT法检测材料对L929细胞活性的影响;将L929细胞接种于被覆PLLA/PLLA-gHAP材料的盖玻片表面上,于1、2和3 d用倒置显微镜观察细胞形态学改变;将PLLA/PLLA-gHAP材料浸提液对小鼠进行腹腔内注射,观察小鼠的急性毒性反应,计数鼠骨髓多染红细胞微核率以检测遗传毒性作用;将PLLA/PLLA-gHAP板植入兔皮下,于2周、3个月和6个月检测兔血液学指标改变,光镜观察植入物周围组织病理学变化。结果:PLLA/PLLA-gHAP材料浸提液对L929细胞的增殖率(RGR)和细胞毒性分级(CTS)的影响与阴性对照组比较差异无显著性(P>0.05),细胞在材料表面生长良好;材料浸提液对小鼠无急性毒性及遗传毒性(微核率0.24%);材料在动物体内埋植后,早期组织内有淋巴细胞、巨噬细胞浸润,纤维膜形成,6个月时纤维膜基本消失,炎症反应明显减轻。结论:PLLA/PLLA-gHAP材料具有良好的生物相容性。Objective To investigate the biocompatibility of poly (L-lactide) /poly (L-lactide) surface grafted hydroxyapatite composite (PLLA/PLLA-gHAP) as a new biodegradable material, and provide a biological evidence for the application of the material. Methods The L929 cell bioactivities in PLLA/PLLA-gHAP group, PLLA group, negative control group (DMEM culture media), positive control group (DMEM contain phenol solution) were determined by MTT assay after cocuhure for 24, 48 and 72 h. L929 cells were cultivated on cover glide coated with PLLA/PLLA-gHAP composite for 1, 2 and 3 d, phase contrast microscope was used to observe the morphological changes of the cells. The leaching liquor of the material was injected into mice to test the acute toxicity and genetic toxicity of it. The PLLA/PLLA-gHAP plates were implanted subcutaneously into rabbit, and the anatomical, pathological and hematological changes were examined after 2 weeks, 3 months and 6 months. Results There was no significant difference in cytotoxicity between the PLLA/PLLArgHAP goup and negative control group, and the L929 cells grew well on the cover glide which goated with PLLA/PLLA-gHAP composite. No any acute toxicity or genetic toxicity was found in mice. There was only mild inflammatory reaction and thin fibrosis membrane in the early days after PLLA/PLLA-gHAP plate implantation, the fibrosis membrane was nearly disappeared and most of the imflammatory reaction recovered 6 months later. Conclusion The new bioabsorbable material, PLLA/PLLA-gHAP composite, has good biocompatibility .
