摘  要:目的:探讨氟尿嘧啶(5-Fu)联用丝裂霉素(MMC)治疗大肠癌体外发生协同作用或拮抗作用时基因群的表达差异。方法:用TRIzol法抽提5-Fu+MMC体外存在拮抗作用和协同作用的肠癌组织的总RNA,反转录荧光标记cDNA探针,与人17K基因表达谱cDNA芯片杂交,经严格洗片后扫描芯片荧光信号图像,计算机分析后比较两种组织中差异表达的基因。结果:在17 101条基因中,5-Fu+MMC体外拮抗作用和协同作用的肠癌组织之间存在差异表达的基因共300条(1.8%),其中上调240条(1.4%),下调60条(0.4%)。表达异常的基因与细胞染色质组蛋白的组成,细胞增殖、转化、癌变,人体免疫功能等相关。结论:微矩阵基因芯片技术具有快速、高通量、高敏感度等特点,结肠癌5-Fu+MMC体外存在拮抗作用和协同作用的基因差异表达谱的分析为揭示其分子机制提供了新的思路与线索。ORIECIIVE To search for the differentially expressed genes between synergism and antagonism of 5-Fu + MMC in colon cancers in vitro. METItODS The RNAs were isolated from colon cancers in which synergism and antagonism were present in vitro of 5-Fu + MMC using TRIzol method, and reversely transcribed to the cDNAs with the fluorescent probes, then hybridized to the human 17K cDNA microarray. After high stringent washing, the cDNA microarray was scanned for fluorescent signals and analyzed for different expression of the target genes between the two tissues. RESULTS Among the 17 101 genes, 300 genes(1.8%) were differentially expressed between the two tissues, including 240 genes up-regulated(1.4%) and 60 down-regulated(0. 4% ). The differentially expressed genes were associated with histones of chromosomal fiber, cell proliferation, transformation, cancerous, immune function. CONCLUSION cDNA microarray technique is effective, rapid, sensitive. The analysis of screening differentially expressed genes between in vitro chemosensitivity of 5-Fu plus MMC in colon cancers provided new clues in demonstrating its molecule mechanism.
