摘  要:目的动态观察虫草菌丝提取物(CJTW)对二甲基亚硝胺(DMN)大鼠肝纤维化和脂质过氧化损伤的影响,探讨其作用机制。方法采用DMN 4周12次腹腔注射制作大鼠肝纤维化模型,自造模之日起,模型大鼠经口给予虫草提取液,分别于给药后3天、2周及4周动态观察药物的干预作用。结果(1)模型大鼠肝组织病理染色,肝组织羟脯氨酸(Hyp)含量测定、血清透明质酸(HA)浓度均显示成功建立DMN诱导大鼠肝纤维化模型。(2)与正常组相比较,DMN造模3天后,肝组织H2O2、丙二醛(MDA)的含量即升高,而后随着造模的进行继续升高,4周时达到峰值。(3)虫草提取物显著抑制模型3天时H2O2含量,在肝纤维化形成中,虫草提取物能改善肝功能和肝脏病理损伤,减轻肝组织中胶原纤维沉积;模型4周时血清丙氨酸转移酶(ALT)、门冬氨酸氨基转移酶(AST)活性、HA含量和肝组织H2O2、MDA、HYP含量通过虫草提取物的干预得到显著的抑制。结论CJTW具有抗DMN大鼠肝纤维化的作用,抗过氧化损伤是其重要机制。Objective To observe the effect of Cordyceps sinensis extract on liver fibrosis and lipid pemxidation induced by Dimethylnirosamin(DMN) and investigate the mechanism.Methods Liver fibrosis model in rats was duplicated by intraperitoneal injection of DMN for 4 weeks and 12 times.At the beginning of modeling, rats were administrated with cordyceps sinensis extract orally to observe inhibiting effect of cordyceps sinermis extracton the model dynamically at 3rd day, 2nd week and 4th week respectively. Results (1) DMN induced liver fibrosis in rats was successfully established by pathostaining,liver tissue hydroxyproline(HA) and serum hyaluronic acid(Hyp) determination; (2)Comparing with the control group, H2O2 content continuously increased after 3 clays and peaked at fourth week of modeling. (3) Cordyceps sinensis extract significantly decreased H2O2 content at 3 day of modeling, it improved liver function,reduced liver injury and collagen deposition during the formation of liver fibrosis, it was also able to inhibit the increase of ALT,AST activity,HA content in serum as well as H2O2 ,MDA and HYP content in tissue at fourth week of modeling. Conclusion Cordyceps sinensis extract have the inhibited effect on liver fibrosis induced by DMN and inhibition of oxidative injury is an important mechanism for it.
