摘  要:目的 用不同接种体积立体定向建立大鼠C6脑胶质瘤模型,比较其生长特性。方法 配制浓度为1.0×10^6个/10μL C6单细胞悬液,分别取5、10、20、40、80、160μL立体定向接种于大鼠右侧大脑尾状核区,在四个水平对各组进行比较。结果实验大鼠成瘤率分别为60%(5μ/L)、93%(10μL)、100%(20μL)、93%(40μL)、87%(80μL)、33%(160μL),瘤细胞病理性核分裂像多见,C-erbB1和S-100B等神经胶质瘤常见蛋白强阳性表达。结论 建立大鼠C6脑胶质瘤模型,接种体积在10-80μL时,成瘤率高,颅内肿瘤生长稳定,肿瘤组织形态学及病理学特性与人脑胶质瘤相似,而且实验周期短、可靠性高,是临床胶质瘤基础研究的理想模型。Objective To establish brain glioma models in rats with different-volume C6 cell suspension by stereotaxic implantation and to compare the growth patterns of the intracranial tumors. Methods We firstly prepare 1.0×10^6/10μL C6 cell suspension, implant it stereotaxically into the right caudate nucleus at different volumes (5, 10, 20, 40, 80, 160μL), and then observe the living states of rats, investigate the volumes, metastasis, histopathology, expressions of C-erbB1 and S-100B proteins of tumors. Results The formation rates of brain tumors were 60%(5 μL), 93%(10μL), 100%(20μL), 93%(40μL), 87%(80μL), 33%(160 μL). The tumors were highly cacoethic, with remarkable nuclear polymorphism. Immunohistochemical staining showed C-erbB1 and S-100B proteins were highly expressed in rat tumors. Conclusion The rat C6 glioma models can be successfully established with higher tumor formation when the implantation volume was from 10μL to 80 μL. The rat C6 glioma resembles human glioma in histopathological and morphological features, so it can be used as an ideal model for studying the glioma.
