摘 要:目的:探讨不同离心方法制备的富血小板血浆(platelet-richplasma,PRP)对牵张成骨的影响,确定理想的PRP制备方法和浓度。方法:将36只健康白兔随机分为3组,每组12只。选取3种方法制备PRP,于牵张期注射至各组的牵张间隙中。牵张结束后2、4、8周每组各处死4只动物取材。双能量X线骨密度仪(dualenergyX-rayabsorptionmetry,DEXA)测量新生骨骨密度,HE染色观察新骨生成情况。结果:注射Landesberg法制备的PRP组新骨生成情况优于其余2组,骨密度值有显著性差异(P<0.05);注射Anitua法和Aghaloo法制备的PRP组新骨骨密度值无显著性差异(P>0.05)。结论:Landesberg法制备的PRP对牵张成骨中新骨生成具有显著促进作用;2次离心法,以200×g力离心10min制备的PRP,血小板浓度为全血的500%左右为最佳。Objective: This study is to evaluate the effect of platelet-rich plasma (PRP) with different centrifugal methods to find out the best method and concentration.Method: Thirty-six white rabbits were randomly divided into three groups. Three methods with different centrifugal times,force and time were selected to make PRP. The three groups were injected PRP at lengthening period, respectively. Four animals from each group were sacrificed at 2,4, 8 weeks at the end of distraction.Dual energy X-ray absorption metry (DEXA) was used to measure regeneration bone mineral density (BMD), HE coloration was utilized to observe bone regeneration.Result: Bone formation rate of the group were injected PRP with Landesberg method was better than injected PRP with the others methods, there were significant differences in BMD (P〈 0.05). There were no significant difference in BMD between the groups were injected PRP with Anitua method and Aghaloo method (P〉0.05).Condusion:Landesberg method of making PRP has an significant accelerative effect during bone distraction.Twice centrifugation with 200×g force , 10 minutes and about 500%concentration was the best.
