摘 要:目的探讨榄香烯(Ele)和姜黄素(Cur)对体外培养的牛晶状体上皮细胞(LEC)凋亡的诱导效应及其机制。方法将培养的牛LEC分别与160μg/mL Ele和20μg/mL Cur在CO2培养箱中共同孵育8,16,24,48,72 h,通过透射电子显微镜观察LEC超微结构;采用流式细胞术(FCM)测定LEC的DNA含量及线粒体跨膜电位(ΔΨm)。结果LEC分别与Ele和Cur共同孵育后,电子显微镜下可观察到细胞核染色质凝集、固缩、边集、核碎裂等,均呈现典型的细胞凋亡的形态学改变。FCM分析可见典型的凋亡亚G1峰,提示细胞核内DNA含量下降,并随药物作用时间的延长而加强。FCM检测发现细胞质出现线粒体ΔΨm下降,且在药物作用早期已发生变化。结论Ele和Cur能显著诱导LEC凋亡;细胞核内DNA含量下降、细胞质内线粒体ΔΨm降低,分别是Ele和Cur诱导LEC凋亡的细胞核和细胞质途径。线粒体ΔΨm下降是Ele和Cur诱导LEC凋亡的早期事件。Objective To investigate the effect and the mechanism of elemene(Ele) and Curcumin (Cur) on apoptosis of lens epithelial cell(LEC) in vitro. Methods The bovine LEC cultured with 160 μg/mL Ele and 20μg/mL Cur were observed by using transmission electron microscopy. The changes of DNA content and mitochondrial transmembrane potential(ΔΨm) in LEC were detected by flow cytometry. Results The typical morphological changes of LEC apoptosis in Ele and Cur group were observed under transmission electron microscope, such as chromatin condensation and aggregation at the nuclear periphery, and nuclear fragmentation as well. The DNA content of LEC in Ele and Cur group decreased time- dependently. The ΔΨm of LEC in Ele and Cur group decreased from early stage. Conclusion Ele and Cur can remarkably induce apoptosis of LEC in vitro. The decrease of DNA content in LEC nucleus and the collapse of mitochondrial transmembrane potential(ΔΨm) in cytoplasm are the nucleu and cytoplasm events of apoptosis mechanisms. The decreased of ΔΨm induced by Ele and Cur is the early event of LEC apoptosis.
