摘  要:根据GenBank上发表的牛α干扰素基因序列,设计了一对特异性引物。提取黄牛血液中白细胞基因组DNA,PCR扩增出α干扰素基因,并与pMD18-T克隆载体相连。测序结果表明,扩增片段含有498个核苷酸,编码166个氨基酸的成熟蛋白,与已报道的牛α干扰素C亚型最相近,氨基酸序列同源性为97.6%。获得BoIFNα-成熟蛋白基因,为重组牛干扰素的开发奠定了基础。One pair of specific primers of bovine interferon α gene was designed and synthesized according to the published sequence in GenBank. Interferon α gene was cloned from genomie DNA of Chinese Luxi yellow cattle by PCR, and the PCR product was inserted into vector pMD18-T to construct plasmid pMD18-T/BoIFN-α. Sequence analysis showed that the Chinese Luxi yellow cattle IFN-α gene was composed of 498 nueleotides, eneoding a mature polypeptide with 166 amino aeids. Compared with other BoIFN-α subtypes, the amino aeid homology was 97.6 %. The results showed that the eloning plasmid was suecessfully eonstrueted and it laid a foundation for the produetion and development of Luxi yellow cattle interferon α gene.
