丙泊酚的遗忘作用及其与γ-氨基丁酸A受体的关系

摘　　要：目的观察丙泊酚对小鼠学习记忆功能的影响及其与γ-氨基丁酸A(GABAA)受体的关系。方法按分层随机区组设计将小鼠分成5组(n=10):脂肪乳10ml·kg-1组(Ⅰ组),丙泊酚25mg·kg-1+生理盐水10ml·kg-1组(P25+NS组),丙泊酚25mg·kg-1+一叶秋碱1mg·kg-1组(P25+Se1组),丙泊酚25mg·kg-1+一叶秋碱2mg·kg-1组(P25+Se2组),丙泊酚25mg·kg-1+一叶秋碱4mg·kg-1组(P25+Se4组)。用避暗实验、跳台实验测定给药后24h小鼠的潜伏期及错误次数。结果与P25+NS组相比,P25+Se1组的潜伏期和错误次数与之相似(P>0·05),P25+Se2组与P25+Se4组潜伏期延长(P<0.01),错误次数减少(P<0.01),似呈剂量依赖性,但不能恢复至Ⅰ组水平(P<0.01)。结论GABAA受体部分介导了丙泊酚的遗忘作用。Objective To investigate the relationship between GABAA receptor and the amnesic effect of propofol. Methods The mice were randomly divided into 5 groups （n = 10 each） ： Ⅰ group （intralipid 10 ml ·kg^-1 group）, P25 ＋NS group （propofol 25 mg·kg^-1 ＋normal saline 10 ml · kg^-1 group）, P25 ＋Se1 group （propofol 25 mg·kg^-1 ＋securinine 1 mg · kg^- 1 group）, P25 ＋ Se2 group （ propofol 25 mg·kg^-1 ＋ securinine 2 mg·kg^-1 group）, and P25 ＋ Se4 group （propofol 25 mg ·kg^-1 ＋ securinine 4 mg·kg^-1 group）. The step- through/step- down latency and the number of errors were recorded 24 h after the drugs were given in the tests. Results As the P25 ＋ Se2 group and the P25 ＋ Se4 group were compared with the P25 ＋ NS group, the step - through latency and the step - through latency prolonged and the numbers of errors decreased（ P 〈 0.01 ） ,seemingly in a dose- dependent manner; but none of them attained the levels of Ⅰ group. Conclusion GABAA receptor partially mediates the propofol - induced amnesia in mice.
