中药复方对糖基化终末产物培养下的肾小球系膜细胞增殖以及系膜基质增生的影响

摘　　要：目的观察中药复方对糖基化终末产物(AGEs)培养下的肾小球系膜细胞增殖以及细胞外基质分泌的影响。方法MTT法检测肾小球系膜细胞的增殖,碱解法检测细胞上清液中羟脯氨酸的含量。结果1×10-3、1×10-2、1×10-1mg/mL的中药复方提取液均能抑制AGEs培养下的鼠肾小球系膜细胞的增殖;能减少AGEs作用下的细胞上清液中羟脯氨酸含量,抑制系膜基质增生。结论中药复方能抑制AGEs引起的大鼠肾小球系膜细胞增殖以及系膜基质增生,提示此方可能通过阻止AGEs受体信号通路抑制糖尿病肾病的发生发展。Objective To observe the effect of the prescription on the progress of mesangial cell proliferation and matrix secreted induced by AGEs. Method The proliferation of glomerular mesangial cell cultured in media was measured by MTT. Hydroxyproline level was measured by alkaline hydrolysis. The proliferation of mesangial cells were obviously increased by AGEs （0.25 mg/mL）. The proliferation mesangian cells and the hydroxyproline levels were suppressed by the prescription of the concentration 1 × 10^-3, 1 × 10^-2, 1 × 10^-1 mg/mL. Conclusion The prescription could suppress the proliferation of mesangial cells and extracellular matrix induced by AGEs, It suggested that the prescription play a significant role in the prevention of diabetic nephropathy by inhibiting the AGEs receptor.
