摘  要:目的探讨P选择素(selectin)在大鼠胰腺移植缺血再灌注损伤中的作用。方法将45只SD大鼠随机分为假手术组(15只)和全胰十二指肠移植组(15对),于再灌注后1、3、6h取血测定血清淀粉酶水平,并切取胰腺标本进行组织病理学观察及P选择素免疫组化染色。结果移植组胰腺组织损伤随再灌注时间的延长而加重,血淀粉酶升高,与中性粒细胞浸润直接相关;而假手术组胰腺组织损伤不明显,血淀粉酶不升高。移植组各时段P选择素均表达,且再灌注后1h即达高峰,假手术组P选择素不表达。结论中性粒细胞的活化以及释放出各种蛋白酶和活性因子,是导致胰腺移植缺血再灌注损伤的直接原因。P选择素在大鼠胰腺移植缺血再灌注损伤的病理过程中起重要作用。Objective To investigate the role of P-selectin during ischemia-reperfusion injury of rat pancreas transplantation. Methods Fourty-five Sprague-Dawley rats were randomized into transplanted group and sham operation group. The transplanted group rats received whole panereaticoduodenal transplantation. Pancreas and blood samples were harvested at 1,3 and 6 h after reperfusion for immunohistoehemistry analysis of P-seleetin, histopathologieal and serum amylase assay. Results (1) The damage of graft pancreas tissue was aggravated gradually and the level of serum amylase was elevated markedly as reperfusion was prolonged, which were mild in the sham operation group. These changes were related to neutrophil infiltration. (2)P-seleetin was expressed in the transplanted group, hut was not in sham operation. The expression of P-seleetin peaked at I h after reperfusion. Conclusion (1) It resulted in isehemia-reperfusion injury in pancreas transplantation that neutrophil was activated and released various kinds of proteolytie ferment and active factors. (2)P-seleetin might play an important role in isehemia-reperfusion injury of rat pancreas transplantation.
