摘 要:①目的 探讨五氯酚(PCP)对人类胎盘碱性磷酸酶(PLAP)的作用,为PCP中毒的防治提供理论依据。②方法 应用比色法分别测定有PCP和无PCP存在下PLAP的酶活力,根据其活力变化求出酶的相对活力;用双倒数作图法,确定PCP对PLAP的抑制类型并求其抑制常数(K_1)。③结果PCP的存在抑制了酶活力。当PCP的浓度在6mmol/L之内,随其浓度的增加而酶活力逐渐降低;当PCP的浓度为6mmol/L时,酶的相对活力为52.4%.PCP是PLAP的非竞争性抑制剂,其K_1为3.49mmol/L.④结论PCP对PLAP有抑制作。Objective To study the effect of pentachlorophenol (PCP) on human placental alkaline phosphatase (PLAP) and provide a theoretical basis for prevention and treatment of human PCP poisoning. Method Colorimetry was used to study the activity of PLAP in the presence and absence of PCP- PLAP relative activity was calculated according to the absorbance varia-tions. The inhibition type and inhibition constant Ki of PCP were determined by using the double reciprocal plot. Result PLAP activity was inhibited by PCP. When PCP concentration was below 6mmol/L, its inhibition effect was positively proportional to its concentration; When PCP concentration was equal to 6mmol/L, the PLAP relative activity was 52. 4% , PCP was a noncompetive inhibitor for PLAP; The inhibition constant Ki was 3. 49mmol/L. Conclusion PCP has a inhibition effect on PLAP.
