摘 要:目的应用蛋白质芯片技术筛选单纯性室间隔缺损血清特异标志物。方法采用病例对照设计,用CM10蛋白质芯片检测56例单纯性室间隔缺损患儿与85例儿科常见病患儿血清,筛选血清差异蛋白。结果有21个蛋白具有统计学差异,其中质荷比6640.784、8571.079、6441.972、8698.804、3322.549、2128.960、2291.255、2055.652、2243.846和2.99.661等10个蛋白在VSD患儿血清中呈高表达,而质荷比2050.946、9306.537、7783.535、9198.666、2143.745、4604.572、7993.109、2824.602、2230.218、2083.311和9360.675等11个蛋白下调。结论蛋白质芯片技术是进行室间隔缺损分子流行病学研究的有效工具,所筛选出的蛋白成分可能是单纯性室间隔缺损的血清标志物。Objectlve To screen serum biomarkers from sera of ventrieular septal defect (VSD) children by using proteinChip. Methods With a case - control design, serum samples from 56 isolated VSD patients and 85 patients with common pediatric diseases were detected with CM10 ProteinChip for the protein biomarkers that were significantly different from that in sera of VSD children and control. Results The contents of 21 proteins were significantly different from that of sera of VSD children and controls. Ten of the 21 proteins with the mass/charge (m/z) of 6640.784, 8571.079, 6441.972, 8698.804, 3322.549, 2128.960, 2291. 255, 2055.652, 2243.846 and 2.99.661 were highly expressed in sera of VSD children, and 11 proteins with m/z 2050.946, 9306.537, 7783.535, 9198.666, 2143.745, 4604.572, 7993.109, 2824.602, 2230.218, 2083.311 and 9360.675 were downregulated in sera of VSD children. Conclusion ProtcinChip technology is a effective tool for molecularly epidemiological study on VSD patients. The scroencd proteins are likely to bc the biomarkers in sera of VSD patients.
