姜黄素对肺癌A549细胞bcl-2、bax表达与生长的影响研究

摘　　要：目的:研究姜黄素对肺癌A549细胞的抑制作用机制。方法:采用MTT法测定不同浓度姜黄素作用于肺癌细胞A549后的吸光值以计算其抑制率,并用半定量逆转录聚合酶链反应法测定其bcl-2、bax表达量。结果:不同浓度姜黄素对肺癌A549细胞均有抑制作用,且呈浓度、时间依赖性,并可升高细胞bax及降低bcl-2的表达量。结论:姜黄素可能通过激活bax及抑制bcl-2的表达,从而抑制肺癌A549细胞的生长。OBJECTIVE： To study the mechanism of the inhibition action of eurcumin on lung adenoearcinoma A549. METHODS： The light absorbing values were determined by MTT for the computing of A549 cell inhibition ratio after the lung adenocarcinoma A549 was subjected to the action of different concentrations, and the bcl - 2, bax expression was determined by semi - fix quantify RT - PCR. RESULTS： Different concentrations of curcumin all showed concentration - dependant and time- dependant inhibition action on lung adenocarcinoma A549 cells, and which could increase the bax expression but decrease bcl- 2 expression. CONCLUSION ： The inhibition action of curcumin on lung cancer A549 cells may be attributed to its activation on bax expression and inhibition on bcl - 2 expression.
