摘 要:目的检测pICln蛋白在不同细胞中的分布。方法制备抗pICln蛋白的合成肽血清,并用Western blot对E1等细胞pICln蛋白的表达及猪和大鼠肾脏细胞中pICln蛋白的分布进行检测。结果E1等8种细胞均在相对分子质量约40000的位置出现免疫反应带。猪肾脏pICln主要分布在可溶性胞质中,大鼠肾脏pICln蛋白主要分布在不溶性的膜结构中。结论pICln蛋白在组织、细胞中普遍表达,为其实施某种功能作用奠定基础。Objective To explore the distribution of plCln protein in various cells. Methods Determine 10 kinds of cells including E1 as well as pig and rat kidney cells for the presence of plCln protein by Western blot with the prepared synthetic peptide serum against plCln protein. Results Each of L, HeLa, HCS, G401, LLC-PK1, HL-60, E1 and ROS cells showed a reaction band with a relative molecular weight of about 40 000 on Western blot profile. In pig kidney cells, plCln protein was mainly distributed in soluble cytoplasm. However,in rat kidney cells,pICln protein was mainly distributed in insoluble membrane structure. Conclusion pICln protein was widely distributed in various tissues and cells. It laid a foundation of giving play to the function of pICln protein.
