摘 要:目的:研究核因子-κB(NF-κB)对大鼠气道平滑肌细胞(ASMC)增殖活性的影响。方法:体外培养ASMCs,采用EMSA法直接检测ASMC中NF-κB的活性,MTT法、细胞周期检测及细胞核增殖抗原(PCNA)的免疫细胞化学染色法观察细胞增殖情况。结果:血小板活化因子刺激后ASMC中NF-κB明显活化,各细胞增殖指标均显著增高(P<0.05),NAC可显著抑制上述效应(P<0.05)。结论:NF-κB的活化能直接参与ASMC的增殖。Objective:To investigate the effect of nuclear factor - kappa B (NF -κB ) on the proliferation of airway smooth muscle cells (ASMC). Methods: NF -κB activity in ASMC was examined by electrophoretic mobility shift assay (EMSA). MTF, flow cytometry and immunohistochemistry for proliferating cell nuclear antigen (PCNA) were used to evaluate ASMC proliferation. Results: Platelet - activating factor (PAF) stimulated the NF -κB activity and the cell proliferation in ASMC. The positive rate of PCNA was significantly different among these groups. N - acety - L - cysteine ( NAC ) markedly inhibited the increased NF -κB activity and ASMC proliferation induced by PAF. Conclusion: NF -κB activation may directly involve in the proliferation of ASMC, suggesting its important role in airway remodeling.
