摘 要:本文提出一种新的优化策略.综合分析表面活性剂浓度、缓冲液pH值、有机添加剂浓度、缓冲溶液浓度及操作电压等实验因素对高效毛细管电泳分离的影响,用多种优化目标函数考察分离度和峰分布均匀性。采用均匀设计法安排实验,用统计学方法筛选和建立目标函数回归方程,在此基础上优化分离条件。以7种巴比妥类药物的高效毛细管电泳分离条件的优化为例,获得满意的分离结果。In this paper a new strategy for systematic optimization of HPCE separations has been developed. It analyses comprehensively thc sodium dodecyl sulfate solution concentration, buffer pH value, buffer concentration, organic solvent concentration and the operational voltage versus the effect of the HPCE separations. Statistic methods are extensively used in this strategy: uniform design is employed to examine the effects of multiple experimental factors and their multiple levels, regression analysis and diagnosis are introduced into optimizations, and optimiza tion functions are statisticalIy screcned out of a set of their definitions. It is an empirical and experimentally grounded approach, and little information about the physical or chemical constants of the solutes is needed for optimization. The strategy has been confirmed by the successful prediction of the separation conditions for seven barbiturates.
