摘 要:目的观察大鼠坐骨神经损伤后近端Schwann细胞(SCs)早期免疫组织化学的变化。方法切断成年Wis-ter大鼠右侧坐骨神经(实验组),切除远侧端及其分支,以近侧端为研究对象,术后1、2、3、4、5、6、7、14天取近侧端(离断平面以近)5 mm,进行S-100蛋白和胶质原纤维酸性蛋白(glial fibrillary acidic protein,GFAP)免疫组织化学染色。左侧坐骨神经保持其完整性作为对照组。结果切断坐骨神经后,近侧端GFAP表达逐渐增强,S-100蛋白表达逐渐减弱。结论周围神经损伤后,近侧端SCs存在表型的变化,非成鞘SCs增多,表明不同类型的轴突信号诱导分化产生的不同类型的SCs,在一定条件下能相互转化,这些改变为周围神经再生提供了细胞学基础。Objective To observe immunohistochemistry changes of proximal SCs after sciatic nerve injuries. Methods The sciatic nerves were incised,the distal stump and its branches of right sciatic nerves(experiment group)were resected, the proximal part (5mm) was examined immunohistochemistrially by S-100 and GFAP at 1,2,3,4,5,6,7,14 days.Results Expression of GFAP increased gradully but one of S-100 decreased. Conclusion There were phenotypic changes of SCs of proximal stump after peripheral nerve injuries,non-myelin forming Schwann cells increased,which indicated different types of axons induced and differentiated into different kinds of SCs,transfoming each other,the changes provided cellular basis for regeneration of peripheral nerves.
