摘  要:目的检测HCV感染者血浆和PBMCS中HCV基因型的相关性,同时研究不同型的HCV与丙型肝炎复发及慢性化的关系。方法应用特异性限制性片段长度多态分析(RFLP)—酶切分型法。结果在82例血浆HCVRNA阳性的病例中Ⅱ型为37例,Ⅲ型为34例,Ⅱ/Ⅲ型为11例。在PBMCs中HCVRNA阳性54例,其中Ⅱ型为38例,Ⅲ型为12例,Ⅱ/Ⅲ型为4例。结论人HCV感染后,不但在血浆中可以检测到HCVRNA,也可以在PBMCS中检测到HCVRNA。同时HCVⅡ型比Ⅲ型更易感染PBMCs,并且HCVⅡ型感染者易出现慢性持续性感染,以至于发展为肝硬化。Objective Subject was to study the relation ofHCV genotype in plasma and PBMCs of virulent hepatitis C patients and the relationships between HCV genotypes and severity of hepatopathy, extensive addiction of HCV and relapse and chronicity of diseases.Methods HCV genotypes were classified by way of restrictive fragment length polymorphism, which was also called restriction endonuclease technique (the method of RFLP).Results Among 82 anti-HCV positive and HCVRNA positive cases in plasma there were 54 HCV RNA positive in PBMCs. HCV genotype in plasma showed 37 cases of type Ⅱ, 34 cases of type Ⅲ and 11 cases of type Ⅱ/Ⅲ co-infection.And the results of HCVgenotype in PBMCs were 38 type Ⅱ ,12 typeⅢ ,4type Ⅱ/Ⅲ co-infection. Conclusion These results verified HCV was not only present in plasma, but also in PBMCs after infecting, whichindicated the extensive addicition of HCV RNA. Genotyping of HCV showed that type Ⅱ of HCV more easily infected PBMCs that type Ⅲ ( P 〈 0.01). Moreover, type Ⅱ was more liable to lead to chronic persisitence of bepatopathy, and made the patient's condition gradually worse and even developed into liver cirrhosis.
