摘 要:目的观察苯乙酸(PA)对胶质瘤C6细胞凋亡和细胞内游离Ca^2+浓度的影响,探讨其作用机制。方法体外培养胶质瘤C6细胞,PA诱导分化后,用透射电镜和AnnexinV/PI标记流式细胞仪检测细胞凋亡,Fluo-3/AM探针激光共聚焦显微镜检测细胞内游离Ca^2+浓度的变化。结果电镜观察到凋亡细胞,PA对C6细胞早期凋亡率无影响,主要增加晚期凋亡率,随PA浓度的增加而增加。PA能明显增加细胞内游离Ca^2+浓度,并且随药物浓度的增加而增加。结论PA能诱导C6细胞凋亡,呈剂量依赖性,PA诱导C6细胞凋亡的机制可能与增加细胞内游离Ca^2+浓度有关。Objective To observe the effect of phenylacetate (PA) on apoptosis and the level of intracellular free calcium in rat C6 glioma cells and to study its mechanism. Methods C6 cells were cultured in vitro. After induction by PA, apoptosis was detected by transmission electron microscope (TEM) and flow cytometry (FCM) with AnnexinV/propidium iodide(PI) . The level of intracellular free calcium was determined by laser con-focal scanning microscopy with calcium flourecent probes-Fluo-3/AM. Results Apoptotic cells were found by TEM. PA had no effect on early apoptosis rate, while late apoptosis rate was remarkably increased by PA and increased with the increase of PA concentration. The level of intracellular free calcium was significantly elevated by PA and increased with the increase of PA concentration. Conclusions PA can induce apoptosis in rat C6 glioma cells, which shows a dose-dependent manner. The mechanism of PA on apoptosis might be related with the increase of intracellular free calcium in C6 glioma cells.
