摘 要:目的探讨神经生长因子及与人胰腺癌神经浸润、淋巴转移和病理类型的关系,以及神经生长因子对人胰腺癌细胞株增殖活性的影响。方法采用免疫组织化学技术,检测69例人胰腺癌组织中β-NGF的表达情况,卡方检验分析β-NGF的阳性在胰腺癌神经浸润、淋巴道转移以及病理分型中的作用。构建pNGF的真核表达载体并鉴定后,采用脂质体Lipofectamine将pcDNA3.0-NGF转染至人胰腺癌细胞株Panc-1。MTT法检测转染后细胞的增殖活性。结果β-NGF可在部分胰腺癌组织中表达,且在神经浸润组和无神经浸润组的表达存在显著差异;NGF阳性表达随着肿瘤恶性程度的增高而增高,且与肿瘤的淋巴转移密切相关。MTT检测转染后的细胞增殖活性增强,与空白对照组和转染空质粒组相比,差异有显著性(P〈0.01),且表现出量效依赖关系。结论^-NGF与胰腺癌的浸润、转移密切相关,可增强其增殖活性;检测胰腺癌组织中pNGF的表达,有望帮助评价胰腺癌的预后。Objective To investigate the role of nerve growth factor (NGF) in development of perineural invasion, lymph node metastasis and pathological type of human pancreatic carcinoma and determine whether cancer cell-derived NGF can enhance human pancreatic carcinoma cell proliferation. Methods The expression of NGF was detected by SP immunohistochemical technique in 69 tissue sections of pancreatic carcinoma. Then Chi-square test was used to analyze the role of NGF positive expression in perineural invasion, lymph node metastasis and pathological type. Human pancreatic carcinoma cell line Panc-1 was transfected with pcDNA3. 0-NGF. In vitro proliferation of the cells was analyzed by MTT assay. Results NGF was expressed in some pancreatic carcinoma tissue. Positive expression of NGF was more frequent in perineural invasion group than in non-perineural invasion group, and the difference was significant. Meanwhile, NGF positive expression was also strongly associated with lymph node metastasis. MTT assay showed that stable transfection of β-NGF in Panc-1 cells resuited in enhanced dose-dependent proliferation of carcinoma cells, and the differences were significant (P〈0.01). Conclusions NGF may enhance the invasion, metastasis and proliferation of pancreatic carcinoma cells. Its detection might be helpful for evaluation of the carcinoma prognosis.
