摘  要:目的观察葡萄糖酸锌(ZnG)、氯化锰(MnCl2)及其联合作用对二氧化硅(SiO2)致中国仓鼠肺成纤维细胞(CHL)间隙连接通讯(GJIC)功能下调的影响,并寻找两者的最佳剂量组合。方法常规培养CHL细胞,按实验分组:生理盐水对照组(阴性对照组);SiO2组(100 mg/L);ZnG组(2.25 mg/L);MnCl2组(1.00 mg/L);SiO2+ZnG组(培养液除含100 mg/L SiO2外,分别含1.00,1.50,2.25 mg/L ZnG);SiO2+MnCl2组(培养液除含100 mg/L SiO2外,分别含0.25,0.50,1.00 mg/L MnCl2);ZnG和MnCl2联合作用组(每组除含SiO2100 mg/L外,采用两因素三水平正交设计,ZnG+MnCl2共设9个浓度组)。培养6 h后通过划痕染料示踪技术(SLDT)观察成纤维细胞通讯连接功能。结果与SiO2组比较,ZnG和MnCl2在一定的浓度下可以减轻SiO2对中国仓鼠肺成纤维细胞通讯连接功能抑制作用(P<0.05);ZnG(1.50 mg/L)与MnCl2(0.50 mg/L)联合作用效果最好。结论ZnG,MnCl2及二者联合在体外可减轻SiO2粉尘对中国仓鼠肺成纤维细胞通讯连接功能的抑制作用。Objective To investigate the effects of zinc gluconate and manganous chloride used solely and together on gap junctional intercellular communication level induced by silica dioxide on Chinese hamster lung cells. Methods Well cultured fibroblast cells were divided into seven groups, they were controls, silicon dioxide, silicon dioxide and zinc gluconate, silicon dioxide and manganous chloride, zinc gluconate and manganous chloride. After 6 hours, gap junctional intercellular communication level was measured through serape loading and dye transfer. Results Zinc glueonate, manganous chloride and zinc glueonate together with manganous chloride showed the effects of moderating GJIC induced by silicon dioxide on Chinese hamster lung cells. The most suitable combination of the two compounds should be dosage of zinc glueonate (1.50 mg/L)with dosage of manganous chloride (0.50 mg/L). Conclusions Zinc gluconate and manganous chloride, when used solely and together, could reduce GJIC induced by silicon dioxide on Chinese hamster lung cells.
