摘 要:目的探讨糖尿病大鼠早期肾组织钙神经蛋白(Cal-cineurin,CaN)的表达及意义。方法大鼠随机分:对照组和糖尿病组。应用链脲佐菌素(65mg/kg)腹腔一次性注射方法建立大鼠糖尿病模型。4周末观察大鼠血糖、肾重/体重与尿白蛋白排泄率(AER)的变化。应用免疫组化与West-ern-blotting印迹检测肾组织CaN蛋白表达。结果糖尿病组大鼠血糖、肾重/体重与AER明显高于对照组。免疫组化显示糖尿病大鼠肾组织CaN蛋白表达较对照组明显增加(P<0.01),Western-blotting印迹显示糖尿病大鼠肾组织CaN蛋白表达较对照组增加2.4倍。结论糖尿病大鼠CaN过度表达表明糖尿病肾病发病过程可能与激活CaN信号转导途径有关。Objective To study the significance. Methods Two groups expression of Caleineurin ( CaN ) in the kidney in early diabetic rats and its were randomly designed: control and diabetic. Diabetes was induced with streptozotocin in Sprague-Dawley rats. Relative kidney weight and 24 hours urinary albumin excretion rate (AER) were measured after 4 weeks. The expression of CaN was determined by immuno- histochemistry and Western-blotting analysis, respectively. Results The AER in diabetic group was markedly elevated than that in control group (P 〈0. 01 ). Immunohistochemistry suggested that the expression of CaN protein in the kidney significantly increased in diabetic rats ( P 〈 0. 01 ), and Western-blotting analysis noted that the expression of CaN protein increased 2. 4 fold in the kidney in diabetic rats. Conclusion The expression of CaN protein increases in early diabetic rats, and CaN might be an important signaling mediator of diabetic nephropathy.
