摘 要:目的探讨幼年大鼠反复惊厥后IL-1β和IL-18的表达及川芎嗪对其表达的影响。方法144只20日龄健康SD大鼠随机分为3组:对照组、惊厥组及川芎嗪干预组。通过三氟乙醚反复吸入(连续6次,每天1次)制作幼鼠惊厥动物模型。RT-PCR方法检测各组动物反复惊厥后6 h、1 d、3 d、7 d脑组织中IL-1β、IL-18 mRNA的表达,ELISA方法检测各时相点脑组织中IL-1β和IL-18蛋白表达,同时观察脑含水量变化。结果川芎嗪干预组各时间点脑组织IL- 1β、IL-18 mRNA和IL-1β、IL-18蛋白的表达较惊厥组显著下调(P<0.05或P<0.01),脑含水量较惊厥组显著降低(P<0.01)。结论川芎嗪对幼鼠惊厥性脑损伤具有保护作用,其机制可能与抑制IL-1β、IL-18 mRNA及其蛋白的异常表达有关。Objective The aim of this study was to investigate the effect of Ligustrazine on expression of IL-1β and IL - 18 mRNA in the infant rats in the brain injury following recurrent seizures. Methods 144 postnatal day 20 SD rats were randomly divided into three groups: the control group, the seizure group and the Ligustrazine treated group, seizures in rats were induced by inhalant flurothyl daily in six consecutive days. Brain tissue was sampled at different time points (6 h, 1 d, 3 d,7 d) in each group after last seizure. The expression of IL-1β and IL - 18 mRNA in the brain tissue were detected by RT - PCR and the expression of IL-1β and IL - 18 proteins were measured by ELISA. At the same time, we observed the water content in rats after recurrent seizures. Results The expression of IL-1β mRNA and protein in seizure group exhibited a biphasic pattern, increased significantly as compared with those in control group at post - seizure 6 h,1 d,7 d ( P 〈0.01 ), but there were no significant difference between the two groups at post - seizure 3 d ( P 〉 0.05 ). The levels of IL - 18 mRNA and IL - 18 protein at post - seizure 6 h,1 d,3 d,7 d in the brain tissue of the rat in seizure group were higher obviously than those in control group(at 6 h, P〈0.05, at 1 d,3 d,7 d, P〈0.01 ). In Ligustrazine treated group, the levels of IL - 18, IL-1β mRNA in the brain tissue and water content of brain tissues at different time points ob- viously lower than those in seizure group ( P 〈 0.05 or P 〈 0.01 ). Conclusions Ligustrazine can effectively decrease brain injuries induced by seizures in the infant rats and the protective mechanism of Ligus- trazine on the developing brain injury of seizures is probably related to suppression of the abnormal levels of IL-1β and IL- 18 mRNA in the brain tissue .
