摘 要:目的观察重症急性胰腺炎(SAP)时G蛋折αi2亚单位Gαi2 mRNA在大鼠胰腺组织的表达变化及血小板活化因子(PAF)拮抗剂银杏苦内酯B(BN52021)的影响,从而探讨SAP发病过程中Gαi2与PAF信号转导途径及BN52021治疗机制的关系。方法雄性Wistar大鼠180只,随机分为阴性对照组(NC组),SAP模型组(SAP组)及BN52021治疗组(BN组),每组再分为6个时相点(1、2、3、6、12、24 h),每个时相点10只。观察各组大鼠胰腺组织病理学变化,采用全自动生化分析仪检测血浆淀粉酶,并以逆转录聚合酶链反应(RT-PCR)动态检测Gαi2 mRNA在各组大鼠胰腺组织的表达水平。结果各组大鼠胰腺组织均检测到了Gαi2 mRNA的表达信号。制模后12 h Gαi2 mRNA表达显著上调(t=-7.213,P=0.000),该上调持续到24 h以后(t=-8.597,P=0.000),BN52021治疗未改变这种上调趋势。结论Gαi2 mRNA在正常大鼠胰腺组织有稳定表达,Gαi2有可能参与了SAP的发病过程,但其与SAP时PAF信号转导通路及BN52021治疗SAP信号转导机制的关系尚不十分明确。Objective To observe the dynamic variation of Gαi2 expression in pancreas tissue, investigate the relationship between Gαi2 and platelet activating factor ( PAF) - mediated signaling pathways during severe acute pancreatitis( SAP), and estimate the treatment mechanism of ginkgolide B ( BN 52021 ). Methods 180 male Wistar rats were randomly divided into 3 groups: normal control group( NC). SAP group( SAP) and BN52021 theatment group( BN). Every group was randomly divided into 6 time units (1,2,3,6,12,24 h), 10 rats in each. Observe the pathobiology change of pancreatic tissue, detect the Serum amylase with Hitachi Clinical Analyzer 7180 and the variation of Gαi2 mRNA expression in pancreatic tissue dynamically by reverse transcription polymerase chain reaction (RT - PCR). Results Gαi2 mRNA signal was detected in all time units of NC groups. After 12h of the SAP model was established the expression of Gαi2 begin to improve significantly (t = -7.213, P = 0. 000 ). And this high level expression could still be observed at 24 h after the SAP model was established ( t = - 8. 597, P = 0. 000). Treatment with BN 52021 had no significant effect on the levels of Gαi2 expression. Conclusion Gαi2 mRNA expressed in normal rat pancreatic tissue steadily. And our findings point towards a potential pathophysiological role of Gαi2 in SAP. But the relationship between Gαi2 and PAF - mediated signaling pathways during SAP is uncertain. The role of Gαi2 in treatment mechanism of BN 52021 is unclear, too.
