摘 要:根据VR2332和CH-1aE基因序列设计一对特异性引物,对广西、广东、海南三省15个猪繁殖与呼吸综合征病毒毒株E基因进行RT-PCR扩增,将扩增出的668bp片段插入pMD18-T载体中,进行序列测定。结果显示,15个毒株的E基因核苷酸序列之间的相似性为84.1%~100%,推导编码的氨基酸序列之间的相似性为81.1%~100%;与VR2332、CH-1a、LV株核苷酸之间的相似性分别为85.4%~98.5%、87.1%~96.2%、62.5%~63.8%,与VR2332、CH-1a、LV株氨基酸之间的相似性分别为83.1%~95.5%、86.1%~95.0%、51.7%~57.2%,表明15个毒株的E基因区域变异较大,均属美洲型。将15个流行毒株与国内外已发表的37株猪繁殖与呼吸综合征病毒相应序列进行比较,建立的遗传进化树表明这15个毒株分别属于3个亚群。推导编码氨基酸的比较结果表明,分别属于不同亚群的不同地区流行的猪繁殖与呼吸综合征病毒毒株在E基因编码蛋白的潜在糖基化位点数目、抗原表位区域和与毒力相关的氨基酸等均存在变异。Molecular diversity of porcine reproductive and respiratory syndrome virus was analyzed by sequence comparison of E gene of the 15 virus isolates from three southern provinces (regions). A pair of primers was designed according to the published nucleotide sequences of VR-2332 and CH-1a and used to amplify complete E gene sequence by RT-PCR from virus RNA template. The PCR product of approximately 668 bp was cloned into pMD 18-T vector. Nucleotide sequences comparison revealed a sequence homology of 84.1%-100 % among all 15 local isolates, with a similarity of 81.1%-100 % in deduced amino acid sequences. These local isolates shared 85.4 %-98.5 %, 87.1%-96.2 % and 62.5 %-63.8 % sequence homology with VR-2332, CH-1a and LV strain respectively, which represents 83.1%-95.5 %, 86.1%-95.0 %, 51.7 %-57.2 % similarity in deduced amino acid, sequences. The results showed significant genetic diversity among PRRSV epidemic strains. All 15 isolates belonged to North American genotype. Phylogenetic tree analysis showed that the 15 strains belonged to three subgroups, with diversities evident on potential glycosylation sites, antigenic epitope regions and the amino acids associated with PRRSV virulence based on deduced amino acids orE gene.
