摘 要:目的探讨推迟双足鼠建模时间至1月龄的合理性。方法实验采用雌性封闭群SD大鼠,根据建模时机随机分为3天龄组、3周龄组和1月龄组,每组10只(动物死亡后立即补充,保持各组样本数恒定)。大鼠1月龄后采用高笼喂养,饲养箱内饲料及饮用水悬空,悬空高度与大鼠直立身高相当,并根据大鼠站高增长进行动态调整。以近肩关节截肢法建模,3天龄组直接用丝线捆扎,以剪刀横断上肢。3周龄、1月龄大鼠采取上肢环形切口,钝性分离结扎肌肉血管、肱骨离断截除。对比各组双足鼠存活率以及生长至5、6、7、9、11、13周龄时双足鼠日均累计站立时间的差异,评价不同建模时机对双足鼠模拟人类直立姿势效果的影响。结果3天龄组累计4只乳鼠术后自然死亡,死亡率高达14.3%,9只乳鼠遭母鼠吞噬,概率高达32.1%,建模成功率为35.7%。3周龄、1月龄组模型动物存活率分别为58.8%、100%。1月龄组无术后死亡、母鼠噬子、饥饿死亡现象发生。1月龄组在6、7周龄及11、13周龄时,日均累计站立时间与其他两组比较差异无统计学意义;5周龄、9周龄时日均累计站立时间高于其他两组。结论建模时间推迟至1月龄不影响双足鼠模拟人类直立姿势的效果,还可显著提高建模成功率,是更合理的建模时机。Objective To investigate the feasibility of postponing operation time of the bipedal rat model to 1 month old age. Methods Female SD rats were divided into 3 groups in the experiment. Bipedal rat models were constructed by forelimbs amputation close to shoulder joint at 3 days, 3 weeks and 1 month age for individual group. Every group consisted of 10 rats, in case of rat death, new rats would be recruited to make the model number stable. The rats in every group would be raised in high cage when they were 1 month old and the food and the water were suspended at the level equal to the stand height of the rat. The food and water level were adjusted dynamically according to the stand height of the rat. The 3-day group was constructed by ligation and scissoring directly; the 3-week and the 1 month group were constructed by oper- ation using an circular incision around the upper limb, amputating the humerus after ligating the muscular and vascular tissue. Livability and mean total upright posture time of each bipedal rat group on the day of 5, 6, 7, 9, 11, 13 weeks age were counted and compared to evaluate the influence on the upright-posture-simulating effect by different operation times. Results 4 rats in 3-day group died naturally(14.3%), 9 rats were cannibalized by the mother rat (32.1%), only 35.7% were constructed successfully while laboratory animal livabilities of 3 weeks and 1 month age group were 58.8% and 100% individually. No post-operation death, cannibalism and hungry death phenomena happened in 1 month age group. Mean total upright posture time showed no statistical difference between 1 month age group and the other two groups during 6, 7 week and 11, 13 week old, even longer at 5 weeks and 9 weeks old age. Conclusion Postponing operation time to 1 month age has no negative impact on bipedal rat's human upright posture simulating effect. Moreover, it can improve the livability of model construction. 1 month age is a more rational time for bipedal rat model construction.
