外加参照物不应参加中药指纹图谱相似度计算

摘　　要：目的考察外加参照物对中药指纹图谱相似度计算的影响。方法将10批待测样品(其中参照物为外加内标物)的数据导入中药指纹图谱相似度评价系统(similarity estimate system)软件,通过溶剂峰剪切、尾部剪切、参比峰设定、相对保留时间与相对峰面积计算等预处理,应用夹角余弦法分别计算有参照物色谱峰参与计算的相似度与参照物色谱峰不参与计算的相似度。结果有参照物参与计算的图谱及共有模式的相似度均在0.9以下,当去掉外加参照物的影响后,其相似度均在0.9以上,表明外加参照物的色谱峰对中药指纹图谱相似度计算有较明显的影响。结论外加参照物不应参加中药指纹图谱相似度计算。Objective To study the influence of reference on similarity calculation of chromatographic fingerprint of traditional Chinese medicine. Methods The related data of samples were imported into the software of similarity estimate system （developed by Shenyang Pharmaceutical University）, and the similarity was calculated through the included angel cosine arithmetic with or without the reference after the pretreatment, such as eliminating the peak of solvent, deleting the tail of the chromatographic peak, setting the reference peak, calculating the relative retention time and relative dimensions of peak. Results The value of similarity was below 0.9 with the reference in calculated and upon 0.9 without the reference. The chromatographic peak of external reference could cause apparent influence on the similarity calculation. Conclusions The influence of external reference can not be ignored.
