摘 要:目的:观察兔骨折模型应用低强度脉冲超声治疗后骨折愈合过程中,转化生长因子-β(TGF-β)的表达变化,以进一步探讨其促进骨折愈合的机制。方法:将36只成年新西兰兔随机分成6个时间组,每组6只,均造成双侧桡骨骨折的模型,并于术后24h即开始超声治疗,左前肢为治疗侧,右前肢作为对照侧。各实验兔分别于术后第1、2、3、4、5、6周拍X线片并取双侧桡骨骨缺损处骨痂,制成切片,利用免疫组化染色及图像分析测定TGF-β表达强度。结果:X线片观察,治疗侧的愈合程度优于对照侧(P<0.01)。组织学观察,术后4周,治疗侧骨、软骨组织内TGF-β染色达最高,且持续时间较对照侧长。结论:低强度脉冲超声通过影响骨折不同时期TGF-β的表达及含量的变化,促进骨、软骨组织的损伤修复,有效促进骨折愈合。Objective To observe the effects of low-intensity pulsed ultrasound's facilitation on the expression for TGF-β in the process of fracture-heallng. Methods Thirty-six adult healthy New-Zealand rabbits underwent the artificial double radius fracture and were randomly divided into six groups, six rabbits per group. The left radius received LIPUS-treatment with intensity of 40 mW/cm^2 for 20 min daily at 24 hours after the artificial double radius fraeture operation. The right radius reeeived false treatment as eontrol. X-ray imaging and the levels of TGF-β in bony eallus from fraeture area were performed to assess the effeet of LIPUS at the end of the lst,2nd,3rd,4th,5th and 6th week postoperatively. Results X-ray imaging showed eallus formation and bone linking in the experimental side were much earlier than those in the control side,indicating that healing in the experimental side was better than that in the eontrol side (P 〈 0.01). The positive-indexes of TGF-β in bony eallus from fraetures area were signifieantly higher in the experimental side than those in the control side (P 〈 0.05). Conclusion These results suggest that the low intensity pulsed ultrasound ean effeetively aeeelerate the reparation of injuried bone and cartilaginous tissue via the inereased release of TGF-β during three main stages of bone-heallng.
