摘  要:目的探索流感病毒在细胞中培养的适宜条件。方法采用MEK细胞、FRhK-4细胞、MDCK细胞分别接种甲型流感病毒H1N1型、H3N2型和乙型流感病毒疫苗毒株,观察3种毒株在不同培养条件下的细胞病变、血凝滴度。结果3株流感病毒接种细胞在33℃培养72h后出现拉网、圆缩、脱落等现象;FRhK-4细胞和MDCK细胞对流感病毒较敏感。胞龄为48h的细胞,在NaHCO3的终浓度为1.4‰,TPCK-胰酶浓度为1.0μg/ml的病毒维持液培养流感病毒为最适条件。结论寻找到了细胞流感病毒的适宜条件。Objective To search for optimal culture condition of influenza virus in the tender cells. Methods Influenza A (H3N2, H1N1) and B virus were inoculated in MDCK cells, MEK cells and FRhK-4 cells under different culture conditions to observe cytopathic effect (CPE) , hemagglutination titer. Results after incubated at 33℃ for 72 h, cytopathic effect of viral infection in all three kinds of cells was observed, and hemagglutination assay showed MDCK cells and FRhK-4 cells were more sensitive than MEK cells to influenza vi- rus. It is the best culture condition that the cells aged 48 h in maintenance media containing 1.4‰ NaHCO3, 1.0 μg/ml trypsin-TPCK. Conclusion The optimal culture conditions for influenza virus in cells were showed in this study.
