摘 要:目的探讨青风藤醇提取物对大鼠心脏移植急性排斥反应的作用及受体外周血IL-2、IL-10水平的影响。方法采用大鼠同种异位腹腔心脏移植模型,随机分3组,每组10只。醇提组给予青风藤醇提取物2.5ml/kg(相当于29g生药/kg体重),每日1次灌胃;环孢素组给予环孢素A口服液1.5mg/kg,每日1次灌胃;对照组不应用免疫抑制剂。术后第10天取移植心进行病理组织学检查,依据Stanford标准进行排斥程度分级。ELISA检测受体外周血IL-2、IL-10水平。结果排斥程度分级,醇提组(2.8±0.75)级和环孢素组(1.3±0.46)级低于对照组(3.7±0.46)级,有统计学意义(P<0.05);醇提组高于环孢素组。外周血IL-2水平醇提组(84.16±6.47)pg/ml和环孢素组(56.47±6.40)pg/ml低于对照组(94.36±10.14)pg/ml,有统计学意义(P<0.05)。外周血IL-10水平醇提组(80.28±10.56)pg/ml和环孢素组(92.71±9.05)pg/ml高于对照组(56.41±12.13)pg/ml,有统计学意义(P<0.05)。结论青风藤醇提取物有抗心脏移植急性排斥作用;可降低受体外周血IL-2水平,升高IL-10水平。Objective To study the effect of purified Sinomenium acutum on acute rejection of cardiac allografs and serum IL-2,IL-10 in rats.Methods The cardiac allograft rots were divided into three groups:extract group(Sinomenium acutum-treated 2.5mg/kg per day), CsA group (CsA-treated 1 .5mg/kg per day) and control group(no treatment). The pathological grade according to Stanford and IL-2, IL-10 in serum were measured 10 days after operation. Results The pathological grade in extract group(2.8 ± 0.75) and CsA group ( 1.3 ± 0.46 )were significantly lower than that in control group(3.7 ± 0.46) ( P 〈 0.05), and IL-2 levels in extract group (84.16 ± 6.47 pg/ml) and CsA group (56.47 ± 6.40pg/ml) were also significantly lower than that in control group(94.36 ±10.14pg/ml)( P 〈 0.05),but IL-10 levels in extract group (80.28± 10.56pg/ ml) and CsA group (92.71±9.05pg/ml) were significantly higher than that in control group(56.41 ± 12.13 pg/ml) (P 〈 0.05).Conclusion The extract of Sinomenium acutum can inhibit the acute rejection of heart transplantation, decrease the level of IL-2 and increase IL-10 in serum of receptor.
