摘 要:目的探讨新生猪缺氧后心肌连接蛋白Cx43 mRNA和蛋白含量的变化。方法对照组(C组):新生猪6头,未吸入低氧;缺氧组(H组):新生猪6头,吸入低氧(FiO2=0.1)维持1 h。于缺氧结束后5 h,免疫荧光染色检测心肌组织细胞缝隙连接蛋白Cx43蛋白的表达,实时荧光定量逆转录多聚酶链式反应(RT-PCR)检测心肌组织Cx43 mRNA的表达。结果H组心肌组织Cx43蛋白的表达显著低于C组(P<0.01),但两组心肌组织Cx43 mRNA表达差异无统计学意义(P>0.05)。结论缺氧可导致心肌Cx43蛋白的表达或分布发生变化。Objective To observe the protein and mRNA expression of Cx43 in the heart of hypoxic newborn piglets. Methods Twelve anesthetized and mechanically ventilated piglets (5 - 7 days old) were randomly divided into two groups: (1) Group C, not exposed to hypoxia; (2) Group H, inhaled 10% oxygen for 1 h. Five hours after hypoxia, Cx43 expression was revealed by immuno fluorescence staining. Cx43 mRNA was calculated by real-time fluorescence quantitative RT-PCR. Results After hypoxia, protein expression of Cx43 in group H was significantly decreased as compared with that of group C ( P 〈 0.01 ). However, there was no significant difference in the mRNA expression of Cx43 between the two groups ( P 〉 0.05). Conclusions Hypoxia may induce changes of the protein expression or location rather than the transcription of Cx43 in newborn piglets.
