摘  要:[目的]观察小鼠异位气管移植后不同时间点基质金属蛋白酶-9(Matrix metalloproteinase-9,MMP-9)及其抑制剂金属蛋白酶组织抑制因子-1(Tissue inhibitors of metalloproteinase-1,TIMP-1)表达水平的变化,探讨其在肺移植后闭塞性细支气管炎(bronchi olitisobliterans syndrome,BOS)中的重要作用。[方法]建立小鼠异位气管移植模型并于术后7、14和28d取材。HE和mallory染色形态学观察、并检测术后气管闭塞率、免疫组织化学检测气管移植物内α-SMA、MMP-9和TIMP-1的表达情况并测定MMP-9与TIMP-1比值,分别加以比较。[结果]移植后1~14d MMP-9表达显著增强并于14d达峰值,14~28d逐渐降低;1~28d TIMP-1表达不断增强,各时间点均显著大于对照组并于28d达峰。[结论]移植后MMP-9/TIMP-1比例失衡,早期使移植物持续性损伤、细胞外基质反复破坏,后期使移植物修复、重建而纤维化,最终导致BOS不断发生发展。[Objective] To study the differential expressions of MMP-9 and TIMP-1 after marine heterotopic tracheal transplant at different time spots, and explore its contribution to BOS after lung transplantation. [ Methods] The spocimens were drawed from the model of heterotopic tracheal transplant at the day of 7, 14 and 28 after lung transplantation, and stained with HE and mallory. Then morphologic studies was done and immunohistochemical assays were carried out to determine the rate of trachea obliteration, the expressions of α-SMA. MMP-9 and TIMP-1 and MMP-9/TIMP-1 ratio in the tracheal transplants. [Results] The level of MMP-9 increased significantly during 1-14 d and peaked at the day of 14, decreased gradually druing 14-28 d. But the level of TIMP-1 increased persistently and peaked at the day of 28, and was higher significantly than control group at every time spot. [Conclusions] A dynamic unbalance between MMP and "lIMP activity makes transplants durative injuries and extracellular matrix interative injuries in early phase, makes transplants repair, remodel and fibrosis in late phase, and finally results in the development of BOS.
