摘  要:[目的]研究乳鼠心肌细胞CD80/CD86的表达模式并分析该表达模式对同种异体淋巴细胞的功能影响。[方法]取BALB/c(H-2d)和C57BL/6(H-2b)乳鼠心脏,分离培养心肌细胞,用RT-PCR及免疫荧光测CD80/CD86表达。将培养10d乳鼠心肌细胞与预活化CTL共培养4h,同时另设CD80或CTLA-4封闭组,计算CTL杀伤率。[结果]培养的乳鼠心肌细胞表达CD80,不表达CD86。单独表达CD80心肌细胞通过CD80与CTLA-4结合减弱CTL杀伤。[结论]培养的乳鼠心肌细胞单独表达CD80共刺激分子能有效抑制CTL杀伤。[ Objective] To investigate the expression pattern of CD80/CD86 on neonatal mice cardiomyocytes and how the cells work when they meet aUogenic lymphocytes. [Methods] Cardiomyocytes were isolated from neonatal BALB/c or C57 mice and cultured for 10 days, then stained with CD80/CD86 antibodies and analyzed by immunofluorescence. CD80/CD86 mRNA were also analyzed by RT-PCR. Effector CTLs were C57 splenocytes which were incubated with stimulator cells (BALB/ c splenocytes) for 2 weeks to yield matured CTL. Target cells were cultured BALB/c cardiomyocytes. [ Results] CD80, but not CD86 expressed on cardiomyocytes after 10 days culture and the cells attenuated the CTL-mediate lysis through the combi- nation of CD80 and CTLA-4. [Conclusions] The neonatal mice cardiomyocytes expressing CD80 costimulator exclusively can inhibit the killing of CTL effectively.
