摘 要:[目的]探索新分离到的甲1亚型(H1N1)流感病毒血凝素(HA)基因特性。[方法]鸡胚、传代狗肾细胞(MDCK)分离流感病毒;血凝抑制试验鉴定型别。逆转录聚合酶链反应(RT-PCR)扩增血凝素重链区(HA1)基因片断,产物纯化后双脱氧链末端终止法测定基因序列,并与1977年来的6个国际、国内代表株比较。[结果]抗原性分析:新分离的H1N1株与国家代表株A1/北京/262/95有大变异(64)。基因分析:HA1区有104个核苷酸的替换或缺失,导致40个氨基酸的改变,涉及到4个重要抗原决定簇区。新分离株与代表毒株相比核苷酸、氨基酸同源性逐年增加。[结论]分子生物学和血清学研究证实1997年河北省出现了新变异株,并在人群中造成广泛流行。[ Objective] To study the regular pattern of types and gene mutation about the influenza viruses isolated rencently. [Methods] Clinical specimens were inoculated in embryonated eggs and MDCK cells, the type of new isolates were identified by hemagghitinin inhibition (HI) test. The viral RNA was extracted by the routine phenol-chloroform method and copied by a pair of oligenucleotide primers specified for H1 through reverse transeription-polymerase chain reaction (RTPCR) , the nucleotide sequence of HA1 genes product were determined by dideoxynucleotide chain termination method and compared with that of 6 representative strains published since 1977. [ Results] Antigenic analyses indicted that the new isolates were greatly different from A1/Beijing/262/95 (H1N1) .The nucleotide sequence analyses about HA1 showed that altogether 104 nucleotides were changed or deletion, which only led 40 amino acid changes.11 of the 40 amino acid changes were in 4 antigenic determinants of HA1 protein. [Conclusions] HA1 gene and antigenic analyses reveals that a new variant appeared in Hebei during 1997-1998 influenza season.
