摘 要:测定不同含水量、冷解冻方式处理的苦槠离体胚内源SOD和α-淀粉酶活性,研究其超低温保存效果.结果表明:不同处理苦槠离体胚的SOD和α-淀粉酶活性差异显著,含水量水平是影响其液氮保存的关键因素,而冷解冻方式也是一重要因素.12%含水量和缓冻快解法处理,苦槠离体胚的超低温保存效果最好,SOD和α-淀粉酶活性最高,分别达212.77 U/h和4.764 mg/m in.The activities of endogenous SOD and α - amylase of excised embryos of Castanopsis sclerophylla preserved with different moisture content, freezing and thawing methods were measured and the cryopreservation effects under different conditions were studied. The results showed that there were significant differences in activities of both endogenous SOD and α - amylase under different treatments. It was showed that the moisture content was the key factor to influence the cryopreservation effect for excised embryos of Castanopsis kawakamii and the freezing and thawing methods were other important factors. It was indicated that the best cryopreservation effect could be realized under conditions with 12% moisture content, mild freezing and quick thawing methods, highest activity of endogenous SOD could reach 212.77 U/h and highest activity of α - amylase could reach 4.764 mg/min respectively.
