大鼠空间辨别性学习记忆活动对海马结构Ephrin-A3表达的影响

摘  要:为了探讨大鼠获得空间辨别性学习记忆活动对导向分子ephrin-A3表达的影响,本研究采用免疫组织化学染色和Western blot方法对模型组、游水组和对照组大鼠海马内ephrin-A3的表达部位和表达量进行对比研究。免疫组织化学染色结果显示模型组、游水组和对照组海马结构内各亚区和各层内均可见ephrin-A3样免疫阳性产物分布,模型组大鼠海马CA3区辐射层和腔隙层尤为明显;Western blot方法检测模型组大鼠海马结构内ephrin-A3阳性产物平均积分光密度值明显高于游水组和对照组。模型组内训练14d的大鼠海马结构内ephrin-A3阳性产物平均积分光密度值高于训练7d和21d。以上研究结果提示eph-rin-A3的表达与大鼠空间辨别性学习记忆活动有密切关系。To explore the effect of the spatial discriminative learning and memory on expressing of nervous factor ephrin-A3 in rats, immunohistochemical and Western bloting methods were used to test the quantity difference and positions of ephrin-A3 expression in hippocampal formation in model rats, swimming rats and controll rats. The results showed that positive immunoreactivity products of ephrin-A3 were distributed in every subfields and layers of rat hippocampus formation in model group, swimming group and control group, but those in the stratum radiatum and lacunosum of CA3 of the model group were more obvious. Western blot study showed that significant increase in the IOD of ephrin-A3 positive products could be found in the hippocampal structure of model rats, comparing with that of swimming group and control group. Moreover, the IOD of ephrin-A3 positive products in rats hippocampus on the day 14 was more than that of rats on the day 7 and day 21. The results indicate that there is a correlated relationship between the nervous factor ephrin-A3 and nervous fibers sprouting in hippocampal structure.
