摘 要:利用猪SCAU-LL资源群,根据猪USDA-MARC2.0连锁图谱,选取4号染色体上微卫星标记SW 1678,用变性高效液相色谱(DHPLC)系统和AB I377系统分析了F2代64个个体的微卫星多态性,比较了这2个分析系统在进行微卫星分型中的效率和利弊.2个系统检测到57个样品的基因型相同,有相同的2个样品未检测到产物.另外有3个样品,DHPLC检测出基因型,AB I377未检测到,有2个样品AB I377检测出基因型,而DHPLC未检测到.2个系统检测微卫星多态性的相同率达92.2%,DHPLC未检出率6.3%,AB I377未检出率7.8%,2个系统检测的灵敏度和准确度基本接近,但是DHPLC的检测成本低于AB I377的检测成本.Based on pig linkage map of USDA-MARC2.0, the SW1678 microsatellite marker on SSCA was chosen for genotyping 64 F2 pig samples of the SCAU-LL reference population. The efficiency and deficiency of detecting microsatellite polymorphisms were compared by WAVE nucleotide fragment analysis system (denaturing high performance liquid chromatography, DHPLC) and ABI377 DNA sequencer. Out of 64 individuals, 57 were detected genotype and 2 were missed by both systems, 3 were only detected by DHPLC, and 2 only by ABI377. The results showed the coincidence of detection was 92.2% for the two systems, while failure in detection was 6.3% for DHPLC and 7.8% for ABI377. The sensitivity and accuracy were similar for both systems, but the testing cost was lower for DHPLC than for ABI377.
